Oral route is the common route of administration of analgesia. Oral route is considered as it is easy to be administered by oneself. The main problem of oral route is the fact that it may undergo first pass metabolism which will delayed absorption.
Sublingual route is considered as it may avoid any first pass metabolism and lead to rapid absorption.
Rectal route may also avoid first pass metabolism and rapid absorption as well as requiring smaller dosage. However only a few preparation is available for rectal route and the absorption is erratic.
Intravenous route may produce a rapid effect. However, there is a possibility of toxicity due to overdose. Transdermal route is a form with sustained release and appear to be convenient. Subcutaneous route is a form of continuous infusion which may lead to accumulation and irritation. Intramuscular administration route is a form of sustained release which is painful.
Epidural or spinal route administration is important for surgical analgesia which is requiring a greater degree of expertise.
